該酶從表達(dá)T4 DNA 連接酶基因的大腸桿菌中純化而來，催化雙鏈DNA或RNA的5'-磷酸末端和3'-羥基末端形成磷酸二酯鍵。該酶適用于平末端或粘性末端DNA的連接，修復(fù)雙鏈DNA，RNA，DNA/RNA雜交鏈中的單鏈切口。
